Electroporation-based methods have so far required cells to be in suspension for transfection. However, the 4D-NucleofectorTM Y Unit allows direct Nucleofection of cells in adherence. Cells which typically grow in adherence in culture can be kept and transfected by Nucleofection in their physiological state.
The Primary Cell Optimization 4D-NucleofectorTM Y Kit is suited for rapid determine Nucleofection conditions for primary cell types lacking an Optimized Protocol for the 4D-NucleofectorTM Y Unit. Using one 24-well Dipping Electrode Array and our pmaxGFPTM Vector as positive control, our two Primary Cell NucleofectorTM Solutions for adherent Nucleofection (AD1 and AD2) are tested in combination with a pre-selected set of programs plus controls. The remaining 24-well Dipping Electrode Array can be used for optional fine tuning or performing the first experiment with the selected conditions.
The cells are cultured in the 24-well culture plates provided with the kit*. For the transfection step, a 24-well Dipping Electrode Array is inserted after replacing culture medium by the specific NucleofectorTM Solution mixed with the DNA or RNA substrate.
